BALDWIN PARK, Calif., June 12, 2023 /PRNewswire/ -- As international demand for starting substances for use in cell therapy emerges, California-based regenerative therapeutics company StemCyte announced it has signed a cooperation agreement with a leading US cell therapy company that is developing allogeneic genetically modified CAR-NK cells using umbilical cord blood. StemCyte has agreed to supply the necessary cellular raw materials for allogeneic cell therapy product development worldwide. StemCyte CEO Dr. Tong-Young Lee stated that immunotherapy is currently the main focus of the international cell therapy companies. Since 2017, FDA has approved six autologous CAR-T cell immunotherapy products for cancer treatment. The customized and personalized nature of autologous CAR-T cell therapy involves a very difficult and long manufacturing process, and its high price makes it difficult to promote as a viable first-line cell therapy treatment. In contrast, allogeneic immune cell therapy companies that could carry out large-scale preparation and application could help CAR therapies become more accessible and more acceptable as a treatment option. While there are bottlenecks in scaling up the manufacturing process for allogeneic products, it is difficult to obtain sufficient and stable "raw material" supplies to overcome these challenges to create the industrial value through economies of scale. Umbilical cord blood alone is the most promising source for large-scale production of allogeneic immune cell therapies. Currently, most of the allogeneic immune cell therapy products in clinical development internationally are derived from three sources: adult healthy donors, induced pluripotent stem cells (iPSCs), and umbilical cord blood. To maintain a stable supply of immune cells from healthy donors is an extremely expensive and challenging task. The cell source from a single donor is limited and cannot be used for larger scale. The cell source from a group of healthy donors can lead to concerns about sourcing quality, consistency, and standardization issues in production processes. iPSCs, a new cell source, are facing significant ethical and safety concerns. At present, umbilical cord blood alone is the most promising source for large-scale production and preparation of allogeneic immune cell therapies. Another critical advantage is that all raw materials are collected and stored at the birth of a baby, which is relatively the most stable and safest means of immune cell procurement. In addition to the above-mentioned advantages, StemCyte's RegeneCyte®– a new drug developed using the umbilical cord blood bank resources – has been submitted for FDA approval and is expected to receive approval for licensure this year. The specifications for StemCyte's umbilical cord blood products meet FDA standards while having the experience of more than 2,000 successful transplants globally as evidence. These were key factors in the selection of StemCyte as the primary cord blood supplier for the company's CAR-NK product development. StemCyte, in close partnership with the collection program administrators at UC Davis Health, has increased collections at Southern California hospitals by adding evening shifts at current sites to extend the hours for cord blood collections. Prioritizing areas with high ethnic diversity may ensure better genetic matches for transplant recipients in underrepresented communities. To that end, StemCyte has successfully banked thousands of cord blood units from Latino, Asian and African American communities. Since 2013, 83% of transplant units through the CA-UCBCP have been provided by StemCyte. The renewed contract increases funding to support collection goals to expand public access at designated hospital locations. BALDWIN PARK, Calif., Jan. 26, 2022 /PRNewswire/ -- Southern California cord blood therapeutics company StemCyte Inc. is announcing successful preliminary results for Phase I of the company's clinical trial for treating acute cerebral stroke using monocytes from allogeneic human umbilical cord blood (hUCB). The findings of the Phase I study were published in Cell Transplantation (CLL) in December 2021. A Phase I study was conducted on patients aged 45-80 years who sustained acute ischemic stroke. Umbilical cord blood was obtained from StemCyte's public cord blood inventory based on ABO/Rh blood type, a Human Leukocyte Antigen (HLA) match of > 4/6, and a cell dose of total mononuclear cell (MNC) count of 0.5-5 x 107 cells/kg. Additionally, four (4) 100 mL doses of mannitol were administered intravenously 30 minutes following umbilical cord blood transplantation and every 4 hours thereafter. Primary outcomes were the number of patients who developed graft versus host disease (GVHD) within 100 days after transfusion. Secondary outcomes were changes in the National Institutes of Health Stroke Scale (NIHSS), Barthel index, and Berg Balance Scale scores. In one case, a 46-year-old male patient with a history of hypertension and hemodialysis for end-stage renal disease was treated allogeneically using hUCB with identical ABO/Rh, a 6/6 HLA match, and MNC count of 2.63 x 108 cells/kg. The patient did not present with serious adverse events or GVHD during the 12-month study. His NIHSS score decreased from 9 to 1; the Berg Balance Scale score increased from 0 to 48, and the Barthel index score increased from 0 to 90. This preliminary study showed that an adult patient with hemiplegia due to ischemic stroke completely recovered within 12 months after receiving allogeneic UCB therapy. "We are very pleased with the successful clinical outcome of StemCyte's Phase I study," said StemCyte President and Chairman Jonas Wang, PhD. "With acute stroke being the second and third leading cause of death and disability, respectively, worldwide, this outcome is as outstanding as it was unexpected." Approximately 30%-35% of people who suffer a stroke die and nearly 75% of survivors sustain permanent disability. Current treatments in the acute phase include the use of thrombolytic, anticoagulant, and antiplatelet agents. However, use of such agents increases the incidence of hemorrhage by 15%-20%. Cord blood stem cells proliferate into neural cells, and they were found to be effective in the treatment of several neurodegenerative diseases. In cerebral stroke, the intravenous injection of UCB and MNCs could restore exercise abilities as well as provide neuroprotective effects as indicated by decreased expression of inflammatory markers such as TNF-alpha, IL-1β and IL-2. Most of the Air Force’s RQ-4 Global Hawk high-altitude ISR drones, best known for collecting intelligence over the Middle East and more recently Ukraine, are set to become “Range Hawks”, tasked with monitoring U.S. hypersonic missile tests over the Pacific. Twenty Global Hawks will be converted to Range Hawks at Northrop Grumman’s
Grand Sky facility on Grand Forks AFB, North Dakota. Grand Forks AFB is home to RQ-4s operated by the USAF’s 319th Reconnaissance Wing which continues to fly the most recent Block 40 variant of the remotely-piloted Global Hawks. The Air Force debuted prototypes of the sophisticated high altitude drones in service over Afghanistan shortly after the terrorist attacks of 9-11, 2001. It ultimately acquired a fleet of 32 RQ-4As/Bs in four production Blocks (10/20/30/40) at a total cost of approximately $10 billion. Global Hawk acquisition was plagued by cost overruns over the years. An initial flyaway cost projection of $10 million per aircraft in 1994 ballooned to $131.4 million per RQ-4 by 2013. Though considered an invaluable asset at times, the Air Force appears never to have really loved the Global Hawk. Most of the Block 30 series RQ-4s it is retiring are less than a decade old. Over its career, the Global Hawk’s extended flight endurance and broad area surveillance capabilities (it can survey as much as 40,000 square miles of terrain per day using high-resolution synthetic aperture radar and electro-optical/infrared sensors) have alternately been perceived as too costly, critical to national defense, unreliable, and superior or inferior to the manned U-2. With that reputation in mind, it’s perhaps not a surprise the service has agreed to transfer Global Hawks to DoD’s Test Resource Management Center ( TMRC ) and its “SkyRange” hypersonic missile testing program. SkyRange is, as the name suggests, a framework for aerial test monitoring and data collection, a test range aloft. The concept substitutes airborne Range Hawks with special test sensors for the aging fleet of ships deployed across a Pacific Ocean corridor that TMRC currently uses to test hypersonic missiles. Given the several weeks it takes to deploy and position the ships for each test, it’s only possible to schedule four to six hypersonic missile tests per year. Using the SkyRange Range Hawk fleet, the frequency of tests can theoretically be increased and done so on a nearly impromptu basis. The current test arrangement with sea-based sensors signals the testing schedule to U.S. adversaries. According to TMRC, the modified RQ-4s will also be less expensive to operate and offer the potential of a moveable range, creating additional testing corridors in the Pacific and elsewhere. The Global Hawks join a number of unmanned MQ-9 Reapers (Range Reapers) in the SkyRange fleet. The aircraft will capture real-time telemetry data and multi-spectral full-motion video, while also providing the range clearance and surveillance information, and meteorological data, to support launch decision-making. If the cost and flexibility claims pan out, it’s attractive logic, particularly to the Air Force. Last year, the service announced its intention to retire all of its Block 30 Global Hawks. Air Force Chief of Staff, General C.Q. Brown, claimed in congressional testimony that the unmanned aircraft would be replaced with a mixture of unspecified alternatives, including "penetrating" platforms and "5th- and 6th-generation capabilities." The latter may include the oft-reported on secret stealth spy drone popularly known as the RQ-180 whose purported ISR capabilities appear to be complimented by a communications/data-sharing gateway capability. Written remarks accompanying Gen. Brown’s testimony asserted that legacy ISR platforms like the Global Hawk can no longer survive peer threat environments or deliver needed intel/data fast enough to be relevant. The remarks included the now familiar USAF logic that “These legacy platforms must be phased out, with resources used to invest in modern and relevant systems. Working together, we must take calculated risk now in order to reduce the greater future risk." The Air Force has followed through, transferring the first five Block 30 Global Hawks across the runway from the 319th RW’s hangars at Grand Forks AFB to Northrop-Grumman’s Grand Sky facility last month. The remaining 15 Block 30s will transfer from the 319th to Northrop Grumman by the end of July. In a memo on the transfer Gen. Brown said, “The divestment of this weapons system was a tough but necessary resourcing choice we had to make in order to begin realizing a budgeted savings of over two billion dollars.”

Exactly how the Air Force plans to spend the savings isn’t clear. The service had not answered questions regarding follow-on ISR plans by publication time. General Brown’s emphasis on the $2 billion saved contrasts notably with remarks made by 319th RW commander, Col. Timothy Curry, during the recent transfer. “There’s no way to count how many American and allied lives this specific sensor payload saved between the enhanced integrated sensor suite and airborne signals intelligence,” Curry said. “Putting those capabilities in the hands of our Airmen created near real-time intelligence for warfighters, decision-makers and command centers.”

TMRC consortium awarded second phase of DOE contract targeting production of mixed rare earth oxides from PA coal waste
Texas Mineral Resources Corp. (TMRC), an exploration company targeting the heavy rare earths and a variety of other high-value critical elements and industrial minerals, announced that the Department of Energy’s (DOE) National Energy Technical Laboratory (NETL) has selected a TMRC-led consortium to continue a contract targeting the production of rare earths from Pennsylvania coal waste. This contract option continues the successful completion of the first phase of a DOE solicitation award in September 2020. The award value is approximately $1.1 million. TMRC’s project partners include Penn State, Jeddo Coal Company and McCarl’s. The Texas Mineral Resources consortium objective is to install a self-contained, modular and portable pilot plant at a Jeddo Coal Pennsylvania site, capable of producing 1-3 metric tons of rare earth oxides derived from coal waste material from Pennsylvania anthracite coal. The project commenced 1 October 2020 and resulted in the TMRC consortium successfully concluding a three-month conceptual design phase. This is the fourth US Government award relating to the production of rare earth minerals in which Texas Mineral Resources has participated. In 2016, TMRC announced it had successfully completed a demonstration-of-concept project funded by the US Defense Logistics Agency (DLA) Strategic Materials Division to separate and refine specific high-purity rare earth elements, using a continuous ion exchange (CIX) and continuous ion chromatography (CIC) processing method. In 2019 a consortium including Texas Mineral Resources consortium successfully completed a US Department of Energy Office of Fossil Energy grant to produce multiple separated rare earth minerals from Pennsylvania coal mining waste material. The CIX/CIC method used in both US Government grants is the method being used to process rare earths and additional US Government-listed Critical Minerals from the Round Top (TX) project, being developed by TMRC’s funding and development partner, USA Rare Earth, LLC . Successful completion of the DOE contract is consistent with a commercial supply chain in which final separation of the mixed REE concentrate into individual high purity rare earth oxides would be accomplished at USA Rare Earth’s processing facility using CIX/CIC processing methodology. Shortly after the Phase 2 award announcement, TMRC said that NETL had selected a consortium led by the Pennsylvania State University,  which includes Texas Mineral Resources, for an award targeting critical mineral recoveries from waste streams. The grant, “Carbon Ore, Rare Earths and Critical Minerals (CORE-CM) Initiative for U.S. Basins,” will assess and catalog Northern Appalachian Basin rare earth elements and critical minerals resources and waste streams, develop strategies to recover minerals from these streams, and assess the infrastructure, industries and businesses in the Northern Appalachian Basin to determine supply chain gaps. This is the fifth US Government award relating to the production of rare earth and critical minerals in which Texas Mineral Resources has participated.

Texas Mineral Resources Corp. (TMRC), an exploration company targeting the heavy rare earths, technology metals and a variety of industrial minerals currently through its Round Top Mountain project in Texas, is pleased to announce preliminary geophysical analysis of baseline silver samples taken from the historic Alhambra Mine located in the Black Hawk district of Grant County, New Mexico. In November 2020 TMRC announced the execution of a letter of agreement to pursue, negotiate and subsequently enter into a joint venture agreement with Santa Fe Gold Corp. (OTCQB:SFEG) to jointly explore and develop a targeted silver property within the Black Hawk Mining District in Grant County, New Mexico. Historical exploration activities have demonstrated that the Black Hawk district hosts a well-known geologic type of carbonate vein that contains small but very high-grade ore-shoots or pods containing native silver as the principal ore mineral, with potential for cobalt recovery as well. The nature of these carbonate veins lateral to and between these high-grade ore lenses is not known at this time, but evidence within the Black Hawk district and from historic districts of this type suggests that the intervening vein carbonate is relatively unmineralized. Because of the small size of the ore-shoots, their random distribution, and the lack of definite leads to forecast the proximity of ore, historic attempts to locate and mine these ore-shoots have been unsuccessful. Modern geophysical tools may provide a method of pinpointing these ore-shoots. Geophysical exploration in the mining sector generally employs three methods: gravimetric,  magnetic and electrical. Given the precision of electrical signatures, we believe electrical methods may hold the best promise for locating the Black Hawk District ore-shoots. Optimum conditions for the successful deployment of electro-geophysics would require a strongly responsive target enclosed in an inert wall rock. It is possible to roughly measure these characteristics from rock samples, and representative samples have been analyzed by Zonge International ( http://zonge.com/ ) in their Tucson and Reno labs. One of the principal parameters measured by electro-geophysics is a factor called chargeability:  the ability of the rock to store an electric charge. Chargeability is reported in milliseconds and reflects the time necessary for the rock to return to its static condition after being subjected to the applied voltage. In general, the higher the chargeability number the higher the percentage of metallic minerals. Many other parameters are measured and the complex interactions between them are subject to considerable interpretation and analysis, but a direct comparison of the chargeability gives a good first approximation of the chances of success in locating the target.
