A can­cer cell ther­a­py biotech has reeled in about $20 mil­lion to push for­ward its on­go­ing Phase I.
Al­lo­plex Bio­ther­a­peu­tics has se­cured about $20 mil­lion of an es­ti­mat­ed $25 mil­lion round, per SEC pa­per­work. CEO Frank Bor­riel­lo post­ed to LinkedIn two months ago not­ing the com­pa­ny would look to close a Se­ries A by Jan. 27. Bor­riel­lo, a for­mer VP at Bax­al­ta, formed the US-Aus­tralian biotech in 2016. The biotech’s so-called SU­PLEXA cells, a non-en­gi­neered cell ther­a­py, en­tered a Phase I in Aus­tralia last April for pa­tients with metasta­t­ic sol­id tu­mors and hema­to­log­ic ma­lig­nan­cies, per a US clin­i­cal tri­als data­base. The study is slat­ed to wrap up next Feb­ru­ary and en­roll 60 pa­tients. On Al­lo­plex’s web­site, the com­pa­ny de­scribes its cell ther­a­py as au­tol­o­gous, mean­ing it de­rives from a pa­tient’s own cells, in this case pe­riph­er­al blood com­bined with cel­lu­lar man­u­fac­tur­ing reagents. Al­lo­plex claims it can man­u­fac­ture a full course of treat­ment (4 to 6 week­ly dos­es) in three weeks and re­quires on­ly “stan­dard lab­o­ra­to­ry equip­ment” since they’re not ge­net­i­cal­ly en­gi­neered like ex­ist­ing cell ther­a­pies. — Kyle LaHu­cik

Pair­ing com­pu­ta­tion, bi­ol­o­gy and chem­istry, Hexa­gon says it will de­rive in­sights from fun­gi and bac­te­ria to cre­ate new med­i­cines, ini­tial­ly fo­cused on can­cer and in­fec­tious dis­ease. In con­junc­tion with the fi­nanc­ing, Hexa­gon pro­mot­ed Tara Arved­son to CSO and ap­point­ed Vic­tor Cee as SVP of drug dis­cov­ery. “Mul­ti­ple break­throughs, from peni­cillin to statins to var­i­ous on­col­o­gy drugs, have re­sult­ed from mi­cro­bial nat­ur­al prod­uct drug dis­cov­ery. Bi­cy­cle shares small batch of can­cer tri­al da­ta
Bi­cy­cle Ther­a­peu­tics re­port­ed da­ta from a monother­a­py Phase I dose es­ca­la­tion study of BT8009, its sec­ond pro­gram, at an AS­CO meet­ing in San Fran­cis­co Tues­day morn­ing. At the end of Sep­tem­ber, 49 pa­tients had been dosed in the Phase I por­tion of the Phase I/II tri­al in pa­tients who had re­ceived a me­di­an of three pri­or ther­a­pies. That in­clud­ed 24 pa­tients with urothe­lial can­cer. Out of eight of those pa­tients on the rec­om­mend­ed Phase II week­ly dos­ing, one had a com­plete re­sponse. Three pa­tients par­tial­ly re­spond­ed. As of last month, the me­di­an du­ra­tion of re­sponse was about 14 months, the UK and Boston biotech said, not­ing two of the four re­spon­ders in that group were still on ther­a­py. The biotech said the drug was “well tol­er­at­ed across all 49 pa­tients in the study, with a low in­ci­dence of ad­verse events com­mon to an­ti­body-based ap­proach­es.”
Last fall, the Genen­tech-part­nered com­pa­ny re­port­ed some da­ta from an­oth­er drug, BT5528, show­ing a hand­ful of re­spons­es in a dozen heav­i­ly pre­treat­ed pa­tients with a par­tic­u­lar tu­mor ex­pres­sion. Bi­cy­cle al­so has BT1718, cur­rent­ly be­ing in­ves­ti­gat­ed in a Phase IIa with the help of the Can­cer Re­search UK Cen­tre for Drug De­vel­op­ment. — Kyle LaHu­cik

Disc Medicine has sold shares to raise $62.5 million in a direct offering, it announced Tuesday morning. Of that financing, $50 million came from new investor Bain Capital, followed by existing investors Access Biotechnology and OrbiMed. The Watertown, MA biotech went public at the end of last year via a reverse merger with Gemini Therapeutics, adopting the ticker $IRON to reflect its role as a blood disorders biotech. President and CEO John Quisel told Endpoints News that Disc decided to go public since it has a number of important data readouts this year. “With that comes both the opportunity to recognize value increases in the company, but also the need for capital as we advance deeper into the clinic,” he said. The FDA last week signed off on a safety-related change to the label of Biogen’s controversial Alzheimer’s drug Aduhelm, which has barely mustered any sales since its approval because payers have refused to cover it. The new label makes clear that physicians should, “Inform patients that events of intracerebral hemorrhage greater than 1 cm in diameter have been reported infrequently in patients taking ADUHELM, and that the use of antithrombotic or thrombolytic medications while taking ADUHELM may increase the risk of bleeding in the brain.”

A leader in the protein degradation space, Yale’s Craig Crews, is back with another biotech, Halda Therapeutics. On Tuesday, the startup announced $76 million in funding to take forward a new technology out of Crews’ lab, called RIPTACs , or Regulated Induced Proximity TArgeting Chimeras. The 26-employee startup emerged from his decades-long work on heterobifunctional molecules, he told Endpoints News, dating back to his 2001 paper on PROTACs, which formed the foundation for Arvinas in 2013. With a new deal, Sesen Bio’s merger with CAR-M player Carisma Therapeutics is finally getting the signal to move forward. The deal, announced Tuesday , was reached with an investor group that currently controls 8.7% of Sesen Bio’s shares. The group, led by Bradley Radoff and Michael Torok, had previously called the merger “illogical” and “value-destructive.”
If the merger goes through, Sesen’s spot on Nasdaq would be taken over by Carisma and trade as $CARM . Carisma’s current management team, including CEO Steven Kelly, would lead the combined biotech moving forward. The merger with Carisma was announced back in September after Carisma beat out 41 other companies to merge with Sesen Bio. Sesen said in January that it took four months for its board to evaluate all its options. Read More
A biotech located in Texas’ capital will be looking to enter a SPAC deal. On Monday, the Dallas-based SPAC Financial Strategies Acquisition Corp. announced it will merge with Austin Biosciences Corp., a biotech located in its namesake. Once the deal closes, Austin Biosciences will become a subsidiary of Financial Strategies and will be listed on Nasdaq under the symbol $ASTN . Austin shareholders will receive 12.5 million shares of the acquiring corporation’s stock in exchange for all the capital stock of Austin Biosciences. Financial Strategies said in a release that the value of the combined company will be around $166.3 million, with the current shareholders of the biotech owning around 75% of the shares once the deal finishes. A rare disease outfit in Princeton, NJ just got its paperwork punted back by the FDA. Tiny Soligenix reported Tuesday morning that the FDA handed the company a refusal-to-file letter. Soligenix did not immediately respond to a query from Endpoints News. The letter targeted an NDA submitted for Soligenix’s lead candidate, a synthetic version of hypericin, which is an active component of a plant called Hypericum perforatum, more commonly known as St. John’s wort. The candidate, branded as HyBryte and touting orphan drug and fast track designations from FDA, was submitted to US regulators to be approved to treat early stage cutaneous T cell lymphoma (CTCL). Hub-and-spoke company Replay has launched a new cell therapy company called Syena, co-founded by NK cell leader Katy Rezvani of the University of Texas MD Anderson Cancer Center. Syena plans to put Rezvani’s T cell receptor natural killer, or TCR NK, cell therapies into a Phase I trial in the second quarter of the year. That would make it the first to bring a TCR NK cell therapy into the clinic, Rezvani told Endpoints News. Rezvani added that they’re working on an IND package to submit to the FDA, which should happen “in the next couple of weeks or so.”
